Positional information readout in $Ca^{2+}$ signaling

چکیده
Living cells respond to spatially confined signals. Intracellular signal transmission often involves the release of second messengers like Ca. They will eventually trigger a physiological response, for example, by activating kinases that in turn activate target proteins through phosphorylation. Here, we investigate theoretically how positional information can be accurately read out by protein phosphorylation in spite of rapid second messenger diffusion. We find that accuracy is increased by binding of the kinases to the cell membrane prior to phosphorylation and by increasing the rate of Ca loss from the cell interior. These findings could explain some salient features of conventional protein kinases C.

Positional information, positional error, and readout precision in morphogenesis: a mathematical framework.
The concept of positional information is central to our understanding of how cells determine their location in a multicellular structure and thereby their developmental fates. Nevertheless, positional information has neither been defined mathematically nor quantified in a principled way. Here we provide an information-theoretic definition in the context of developmental gene expression patterns...
متن کاملPKCα: a versatile key for decoding the cellular calcium toolkit
Conventional protein kinases C (cPKCs) play an essential role in signal transduction and are believed to integrate both global Ca(2+) transients and diacylglycerol signals. We provide evidence that PKCalpha is a ubiquitous readout sensor for the cellular Ca(2+) toolkit, including highly restricted elementary Ca(2+) release. Threshold stimulations of cells with Ca(2+)-mobilizing agonists resulte...
متن کاملLimb patterning: from signaling gradients to molecular oscillations.
The developing forelimb is patterned along the proximal-distal and anterior-posterior axes by opposing gradients of retinoic acid and fibroblast growth factors and by graded sonic hedgehog signaling, respectively. However, how coordinated patterning along both axes is accomplished with temporal precision remains unknown. The limb molecular oscillator hairy2 was recently shown to be a direct rea...
متن کاملPositional Relationship between Natural Killer Cells and Distribution of Sympathetic Nerves in Decidualized Mouse Uterus
Background: Uterine natural killer (uNK) cells are the most abundant leukocytes in pre-implantation endometrium and early pregnancy deciduas in humans and rodents. They are associated with structural changes in maternal spiral arteries but regulation of their re-cruitment and activation is incompletely understood. The major subpopulation of uNK cells in humans expresses CD56, the neural cell ad...
متن کاملModel calcium sensors for network homeostasis: sensor and readout parameter analysis from a database of model neuronal networks.
In activity-dependent homeostatic regulation (ADHR) of neuronal and network properties, the intracellular Ca(2+) concentration is a good candidate for sensing activity levels because it is correlated with the electrical activity of the cell. Previous ADHR models, developed with abstract activity sensors for model pyloric neurons and networks of the crustacean stomatogastric ganglion, showed tha...
